Cheltenham mayor Anne Regan cut the ribbon to open the glossy new store.
She had worked for the company for 30 years in various roles.
“I never dreamt I would be opening up the shop as mayor. It is a privilege and I am extremely proud to be here,” she said.
“I pop in and I still know a lot of the staff.”
Marks and Spencers has also helped Mrs Regan get a permanent base for young people off the ground by donating £5,000.
She said: “The firm has been fully behind the community work that I have done so I cannot thank them enough.”
To celebrate the centenary a stylist will be on hand at the High Street shop until Saturday to give people fashion advice.
There will also be a chef in the food hall cooking up seasonal dishes.

“A curious sense of deja vu here...
As I said yesterday, I don't like it.
Cheap-looking tin signs.
The usual race to get all perishables off the shelves to buy in the staff shop before paying customers get them - an hour before closing time.
Talking machines that snap at your heels like a rabid terrier - Edison would be spinning in his grave.
But at least they've had the clock cleaned up. I missed it while it was away.”
