Ian Hutton's Robins side started slowly and found themselves up against it as Truro pressed for an opener.
It arrived in the seventh minute when Helena Calfe's cross from the left was nodded in from close range by Roxanne Cooke.
The home side were lucky not to fall further behind when Sophie Wilson missed a good chance for the Cornish club.
But Cheltenham levelled in the 11th minute and improved steadily as the half wore on.
The equaliser was a well-struck drive from central defender Hazel Adams after a cross from the right by Charlie Griffin fell to her near the edge of the penalty area.
It flew in via a deflection and Griffin tested Truro goalkeeper Katie Pearce with a long-range effort in the 37th minute.
Griffin and Chenise Austin caused problems with their speed, while Reilly enjoyed an impressive debut in attack.
Griffin's created a chance of Austin, but her effort was too high and a fine cross to the far post by Griffin was nodded wide by Donaldson as Cheltenham finished the half in command.
They found themselves behind three minutes after the break however when Wilson rounded Ash Lebbon and applied a low finish into the net from a tight angle.
But Donaldson beat Pearce with a curling shot after a free-kick was touched to her by Griffin. It was awarded for the goalkeeper taking too much time without releasing the ball.
Truro were rattled and Reilly capped her fine performance with the winner, finishing neatly after good work from Donaldson and Austin.
