The restored 1939 sought-after edition of Detective Comics is believed to be one of 150 copies left in the world.
Dominic Winter said: "A perfect copy of this edition sold in the States earlier this year for more than $1 million – a comic world record.
"Our sale is a first for England."
Batman became an instant hit with comic fans and had his own solo title in 1940.
A well-restored edition of this landmark comic creation is also up for auction with an estimated price of £12,000 to £15,000.
Mr Winter said he was expecting a "superhuman buzz" at the sale on December 16.
He said: "Printed paper is our territory, but it is unusual to come across such a collection of comics."
He is hoping to create lots of interest in America. The auction starts from 4.30pm to allow for the time difference.
The comics are part of a 198-strong private collection from a British collector and include rare copies named after many favourite superheroes.
These include a first issue Superman, estimated to fetch up to £15,000, a first edition of the Amazing Spiderman and one of Daredevil, recently named one of Empire's 50 greatest comic book characters.
Mr Winter said: "We are thrilled to see this vast and unique collection of vintage American comics being auctioned. There are some invaluable collectors' editions within the collection that should bring out the inner child and perhaps secret superhero desires in the bidders."
Some of the comics being sold have the highest official grade condition.
