Lowe was recalled to the side in the absence of Sido Jombati, who has undergone double hernia surgery and will miss three weeks of action.
Within three minutes Rotherham were level when winger Courtney Cameron beat Lowe and crossed for Daniel Nardiello to nod in from close range, with Scott Brown unable to deal with the ball into the box.
Cameron was making his debut after arriving on loan from Aston Villa and he caused Cheltenham plenty of problems along with Ben Pringle on the other flank.
Rotherham took the lead in the 20th minute when a cross was headed clear by Alan Bennett, but Pringle controlled it well and beat Russ Penn before burying a firm drive inside the bottom right corner.
A backline that had not been breached in 420 minutes before this afternoon's match was beaten again in the 29th minute.
A simple long ball from the back caught the defence out and Nardiello had time to evade the attentions of Bennett before slotting calmly past Brown.
Cheltenham reduced the arrears two minutes later when Jermaine McGlashan beat the Millers' offside trap and reached a chipped pass from Marlon Pack.
His shot past the advancing Andy Warrington was blocked, but Chris Zebroski was on hand to tap in his fifth goal of the season.
Cheltenham tightened up after the break, but they lacked composure in possession and struggled to retain the ball.
Substitute Jeff Goulding nodded a long throw from Lowe over the bar and a challenge from Ian Sharps denied Shaun Harrad after a fine cross from Sam Deering.
But Warrington was never really tested and midfielder Lee Frecklington added Rotherham's fourth three minutes into stoppage time after a mix-up between Lowe and Deering.
