The Cherry and Whites welcome the Tigers to Kingsholm on the weekend of September 6.
Dean Ryan's men then face a trip to local rivals Bath at the Recreation Ground a week later.
Gloucester close their campaign with a trip to play reigning Premiership champions London Wasps at the Causeway Stadium on the weekend of April 25, 2009.
The Premiership semi-finals and final take place on May 9 and May 16 respectively.
May 16 was chosen to avoid any clash with the opening fixture of the British and Irish Lions tour to South Africa.
Lions tour manager Gerald Davies was grateful for the decision.
“I'm delighted that this issue has been resolved well ahead of the next season thanks to many people's efforts over the last few weeks,” he said.
“I'd like to express my thanks to the English clubs for their gesture in agreeing to stage the Guinness Premiership Final on May 16 and to the RFU for helping to facilitate this. It will allow players to join up with the Lions for the opening match of the tour. ”
